発明の概要(英語) The present invention provides an inactive Ca **2+/calmodulin-dependent protein kinase IIalpha (CaMKIIalpha ) knockin animal in which only the protein kinase activity of CaMKIIalpha has been specifically impaired.
Since CaMKIIalpha is considered to be involved in higher brain functions including learning and memory, and inhibit epileptic seizure and brain disorders caused by ischemia, the inactive CaMKIIalpha knockin animal of the present invention is widely usable for various studies of the brain and nerve, such as the studies of mechanisms of learning disability, dysmnesia, epileptic seizure and brain disorders.
特許請求の範囲(英語) [claim1]
1. An inactive Ca **2+/calmodulin-dependent protein kinase IIalpha (CaMKIIalpha ) knockin nonhuman animal,
wherein a CaMKIIalpha gene of one or both of homologous chromosomes is substituted into an inactive type so that an inactive CaMKIIalpha is expressed, and thereby a protein kinase activity of the CaMKIIalpha is specifically impaired while a calmodulin binding capacity of the CaMKIIalpha and a capacity of multimerizing subunits are maintained; and wherein one or a plurality of amino acid residues in the catalytic domain of the CaMKIIa has been modified.

The inactive CaMKIIalpha knockin nonhuman animal according to claim 1, wherein brain's nucleus accumbens has lower neuronal activity as compared to that of a wild-type, while there is no substantial difference in neuronal activities in the cerebral cortex and corpus striatum as compared to those of a wild-type.

3. The inactive CaMKIIalpha knockin nonhuman animal according to claim 2, wherein the inactive CaMKIIalpha knockin nonhuman animal is produced by a gene targeting method.

4. The inactive CaMKIIalpha knockin nonhuman animal according to claim 3, wherein one or a plurality of amino acid residues that is required for binding to ATP has been modified.

5. The inactive CaMKIIalpha knockin nonhuman animal according to claim 4, wherein a lysine residue that is required for binding to ATP has been modified.

6. The inactive CaMKIIalpha knockin nonhuman animal according to any one of claims 2 to 5, wherein the inactive CaMKIIalpha knockin nonhuman animal is a rodent animal.

7. The inactive CaMKIIalpha knockin nonhuman animal according to claim 6, wherein the inactive CaMKIIalpha knockin nonhuman animal is a mouse.

8. An inactive Ca **2+/calmodulin-dependent protein kinase IIalpha (CaMKIIalpha ) knockin cell,

9. Use of a knockin nonhuman animal in accordance with any of claims 1 to 7 for searching therapy and elucidating pathology of mental disorders, and for drug screening.
