It is found that a transformed plant into which a ³-glutamylcysteine synthetase gene has been introduced is increased in at least one of the number of flowers and the number of seeds in comparison with a wild-type plant, and is therefore increased in seed yield.
Based on this finding, the present invention elucidates the function of a product of a plant-derived ³-glutamylcysteine synthetase gene and provides a technique for producing a plant having an increased seed yield.
特許請求の範囲(英語) [claim1]
1. A transformed plant comprising an expression vector comprising a polynucleotide encoding plant-derived gamma -glutamylcysteine synthetase,
the polynucleotide producing a translated product having no chloroplast targeting signal peptide, the plant being further increased in biomass quantity per unit area or seed yield per unit area in comparison with a parent plant thereof by cultivating the transformed plant planted at a number of individuals per unit area higher than the number of individuals planted per unit area at which the biomass productivity per unit area of a parent plant or wild-type plant of the transformed plant is largest,
wherein said plant further comprises an expression vector comprising a polynucleotide encoding glutathione-binding aldolase, wherein the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (e) to (h): (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity.

2. A method for (A) increasing at least one of the number of flowers of a plant and the number of seeds of the plant or (B) increasing biomass quantity or seed yield of a plant per unit area, the method comprising the steps of: transforming a plant by introducing, into the plant, a polynucleotide encoding plant-derived gamma -glutamylcysteine synthetase; and cultivating the transformed plant planted at a number of individuals per unit area higher than the number of individuals planted per unit area at which the biomass productivity per unit area of a parent plant or wild-type plant of the transformed plant is largest, wherein, in case (A), the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (a) to (h): (a) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 1; (b) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 1, the polypeptide having gamma -glutamylcysteine synthetase activity; (c) a polynucleotide having the base sequence represented by SEQ ID NO: 2; (d) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 2, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity; (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity, wherein, in case (B), (i) a translated product of the polynucleotide encoding the plant-derived gamma -glutamylcysteine synthetase has a chloroplast targeting signal peptide, wherein the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (a) to (d): (a) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 1; (b) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 1, the polypeptide having gamma -glutamylcysteine synthetase activity; (c) a polynucleotide having the base sequence represented by SEQ ID NO: 2; and (d) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 2, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity,
or wherein, in case (B), (ii) a translated product of the polynucleotide encoding the plant-derived gamma -glutamylcysteine synthetase does not have a chloroplast targeting signal peptide, and a harvest index is improved, wherein the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (e) to (h): (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity.

3. The method according to claim 2, wherein, in case (B), further the step of transforming the plant by introducing into the plant a polynucleotide encoding glutathione-binding aldolase is comprised.

A method for producing a transformed plant that is increased in biomass quantity per unit area or seed yield per unit area by cultivating the transformed plant planted at a number of individuals per unit area higher than the number of individuals planted per unit area at which the biomass productivity per unit area of a parent plant or wild-type plant of the transformed plant is largest,
the method comprising the step of: transforming a plant by introducing into the plant a polynucleotide encoding plant-derived gamma -glutamylcysteine synthetase,
and transforming the plant by introducing into the plant a polynucleotide encoding glutathione-binding aldolase, the polynucleotide encoding the gamma -glutamylcysteine synthetase being selected from the group consisting of the following (a) to (h): (a) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 1; (b) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 1, the polypeptide having gamma -glutamylcysteine synthetase activity; (c) a polynucleotide having the base sequence represented by SEQ ID NO: 2; (d) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 2, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity; (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide having at least 97% similarity with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encoding a polypeptide having gamma -glutamylcysteine synthetase activity.

5. A transformed plant into which a polynucleotide encoding plant-derived gamma -glutamylcysteine synthetase is introduced,
the polynucleotide producing a translated product having no chloroplast targeting signal peptide,
the plant being further increased in biomass quantity per unit area or seed yield per unit area in comparison with a parent plant thereof by cultivating the transformed plant planted at a number of individuals per unit area higher than the number of individuals planted per unit area at which the biomass productivity per unit area of a parent plant or wild-type plant of the transformed plant is largest, wherein the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (e) to (h): (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide that hybridizes, under a stringent condition, with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encodes a polypeptide having gamma -glutamylcysteine synthetase activity.

6. The transformed plant according to claim 5, into which a polynucleotide encoding glutathione-binding aldolase is further introduced.

A method for producing the plant according to claim 5, the method comprising the step of: introducing into the plant a polynucleotide encoding plant-derived gamma -glutamylcysteine synthetase, wherein the polynucleotide produces a translated product having no chloroplast targeting signal peptide, wherein the polynucleotide encoding the gamma -glutamylcysteine synthetase is selected from the group consisting of the following (e) to (h): (e) a polynucleotide encoding a polypeptide having the amino-acid sequence represented by SEQ ID NO: 3; (f) a polynucleotide encoding a polypeptide having an amino-acid sequence in which not more than 10 amino acids of which are deleted, substituted, or added from/in/to the amino-acid sequence represented by SEQ ID NO: 3, the polypeptide having gamma -glutamylcysteine synthetase activity; (g) a polynucleotide having the base sequence represented by SEQ ID NO: 4; and (h) a polynucleotide that hybridizes, under a stringent condition, with the polynucleotide having the base sequence represented by SEQ ID NO: 4, and encodes a polypeptide having gamma -glutamylcysteine synthetase activity.

8. The method according to claim 7, into which a polynucleotide encoding glutathione-binding aldolase is further introduced.
