発明の概要(英語) The present invention provides a composition comprising an IP 3 receptor-binding protein (IRBIT), a nucleic acid that controls the expression and translation of IRBIT, or an antibody against IRBIT for controlling at least one intracellular biological function selected from the group consisting of (1) protein synthesis, (2) phosphatidylinositol metabolism, and (3) intracellular pH.

1. Use of inositol 1,4,5-triphosphate (IP 3) receptor-binding protein (IRBIT) for controlling phosphatidylinositol metabolism in a cell in-vitro in which the PIP 2 synthetase isoform alpha , beta or gamma PIPKII (Type II phosphatidylinositol-5-phosphate 4-kinase) is involved, wherein the IRBIT is a protein comprising an amino acid sequence shown in SEQ ID NO: 1 or SEQ ID NO: 3 or a protein comprising an amino acid sequence having 90% or more identity with the amino acid sequence of SEQ ID NO: 1 or SEQ ID NO: 3 and having the same biological activity as IRBIT.

Use of a DNA encoding IRBIT or vector comprising the DNA for controlling phosphatidylinositol metabolism in a cell in-vitro in which the PIP 2 synthetase isoform alpha , beta or gamma PIPKII is involved, wherein the IRBIT is a protein comprising an amino acid sequence shown in SEQ ID NO: 1 or SEQ ID NO: 3 or a protein comprising an amino acid sequence having 90% or more identity with the amino acid sequence of SEQ ID NO: 1 or SEQ ID NO: 3 and having the same biological activity as IRBIT, and wherein the nucleic acid is selected from the group consisting of DNAs encoding IRBIT.

Use of siRNA for IRBIT for controlling phosphatidylinositol metabolism in a cell in vitro in which the PIP 2 synthetase isoform alpha , beta or gamma PIPKII is involved, wherein the IRBIT is a protein comprising an amino acid sequence shown in SEQ ID NO: 1 or SEQ ID NO: 3 or a protein comprising an amino acid sequence having 90% or more identity with the amino acid sequence of SEQ ID NO: 1 or SEQ ID NO: 3 and having the same biological activity as IRBIT, and wherein the siRNA consists of the nucleotide sequence of SEQ ID NO: 5 or 6.

Use of an antibody against IRBIT or a fragment thereof, capable of binding specifically to IRBIT, for controlling phosphatidylinositol metabolism in a cell in vitro in which the PIP 2synthetase isoform alpha , beta or gamma PIPKII is involved, wherein the IRBIT is a protein comprising an amino acid sequence shown in SEQ ID NO: 1 or SEQ ID NO: 3 or a protein comprising an amino acid sequence having 90% or more identity with the amino acid sequence of SEQ ID NO: 1 or SEQ ID NO: 3 and having the same biological activity as IRBIT, and wherein the fragment is selected from the group consisting of Fab, F(ab') 2, Fv and scFv.

5. The use according to any one of claims 1-4, wherein the control is suppression or elevation.

6. The use according to claim 4, wherein the antibody is a monoclonal antibody, a polyclonal antibody, a recombinant antibody, or a single chain antibody.

7. The use according to any one of claims 1 and 4, wherein the IRBIT is phosphorylated.
