発明の概要(英語) Disclosed is a peptide-decomposing reagent having the following properties: 1) the reagent has no significant toxicity such as carcinogenicity; 2) the reagent does not have such excess decomposition properties that any metastable peak can be produced; 3) the reagent does not produce a polyvalent ion peak that is an interference peak; and 4) the separation and sharpness of peaks can be ensured. Also disclosed are: a method for specifically cleaving an N-C.alpha. bond in the main chain of a peptide with the decomposing reagent; and a method for determining the amino acid sequence for a peptide utilizing the specific cleavage. Specifically disclosed are: a method for specifically cleaving an N-C.alpha. bond in the main chain of a peptide, which comprises irradiating the peptide with a laser beam in the presence of 5-aminosalicylic acid; a method for determining the amino acid sequence for a peptide, which comprises irradiating the peptide with a laser beam in the presence of 5-aminosalicylic acid to specifically cleave an N-C.alpha. bond in the main chain of the peptide; a reagent for specifically cleaving an N-C.alpha. bond in the main chain of a peptide; a reagent for releasing a hydrogen radical; a matrix reagent for MALDI-ISD; a matrix for MALDI-ISD; a peptide ionization reagent for MALDI-ISD; and a kit for MALDI-ISD.
