Provided is a method for easily and efficiently producing encapsulated substance vesicles wherein a substance is encapsulated in the cavity of vesicles obtained by polymer self-assembly.
Empty vesicles that have membranes comprising a first polymer that is a block copolymer with uncharged hydrophilic segments and a first kind of charged segments and a second polymer with a second kind of charged segments that carry a charge that is the opposite of said first kind of charged segments as well as spaces that are enclosed by said membranes are mixed in an aqueous medium with the substance that is to be encapsulated in the spaces.
特許請求の範囲(英語) [claim1]
1. (Amended) A process of producing a substance encapsulating vesicle, which encapsulates a substance in a cavity thereof, comprising: mixing a vacant vesicle with the substance in an aqueous medium, said vacant vesicle having a membrane containing a first polymer, which is a block copolymer having an uncharged hydrophilic segment and a first charged segment, and a second polymer, which has a second charged segment charged oppositely to the first charged segment, said membrane defining a cavity surrounded thereby.

2. A process according to claim 1, wherein the mixing is carried out by a process selected from the group consisting of agitating, shaking, and impacting.

3. A process according to claim 1 or 2, wherein the membrane of the vacant vesicle has a trilaminar structure comprising an outer layer, an intermediate layer, and an inner layer.

4. A process according to any one of claims 1 to 3, wherein the substance is selected from the group consisting of biomolecules, organic compounds and inorganic substances.

A process according to any one of claims 1 to 4, wherein the first polymer is a block copolymer containing an uncharged hydrophilic segment and an anionic segment, and the second polymer is a block copolymer containing an uncharged hydrophilic segment and a cationic segment and/or a polymer containing a cationic segment but no uncharged hydrophilic segment.

A process according to any one of claims 1 to 4, wherein the first polymer is a block copolymer containing an uncharged hydrophilic segment and a cationic segment, and the second polymer is a block copolymer containing an uncharged hydrophilic segment and an anionic segment and/or a polymer containing an anionic segment but no uncharged hydrophilic segment.

7. A process according to claim 5 or 6, wherein the anionic segment is nucleic acid.

8. A process according to any one of claims 5 to 7, wherein the cationic segment is polyamine.

9. A process according to any one of claims 5 to 8, wherein the uncharged hydrophilic segment is polyalkylene glycol.

(Amended) A vesicle comprising: a membrane containing a first polymer, which is a block copolymer having an uncharged hydrophilic segment and a first charged segment, and a second polymer, which has a second charged segment charged oppositely to the first charged segment, said membrane defining a cavity surrounded thereby;
and a charged substance encapsulated in the cavity.

11. A vesicle according to claim 10, wherein the membrane has a trilaminar structure comprising an outer layer, an intermediate layer, and an inner layer.

A vesicle according to any one of claims 10 to 13, wherein the first polymer is a block copolymer containing an uncharged hydrophilic segment and a cationic segment, and the second polymer is a block copolymer containing an uncharged hydrophilic segment and an anionic segment and/or a polymer containing an anionic segment but no uncharged hydrophilic segment.

15. A vesicle according to claim 13 or 14, wherein the anionic segment is nucleic acid.

18. A vesicle comprising a membrane containing: a block copolymer having an uncharged hydrophilic segment and a cationic segment;
and a nucleic acid;
said membrane defining a cavity surrounded thereby.

19. A vesicle according to claim 18, wherein the membrane has a trilaminar structure comprising an outer layer, an intermediate layer, and an inner layer.

20. A vesicle according to claim 18 or 19, which has an N+/P ratio of higher than 1.0 and lower than 3.0, the N+/P ratio being a mole ratio of cationic groups of the cationic segment to phosphate groups of the nucleic acid.

21. A vesicle according to any one of claims 18 to 20, wherein the uncharged hydrophilic segment is polyalkylene glycol.

22. A vesicle according to any one of claims 18 to 21, wherein the cationic segment is polyamine.

23. A vesicle according to any one of claims 18 to 22, wherein the membrane further comprises a crosslinker.

24. A vesicle according to claim 23, which has a CL/N ratio of 0.1 or higher, the CL/N ratio being a mole ratio of the cross-linker to cationic groups of the cationic segment.

25. A vesicle according to claim 23 or 24, wherein the cross-linker is glutaraldehyde.

26. A vesicle according to any one of claims 10 to 25, for use in a drug delivery system.
