Abstract In the present invention, a disaccharide unit for synthesizing a useful sugar chain for suppressing the development of dystroglycanopathy is obtained and used to reconstitute a post-phosphoryl sugar chain. Provided are: a disaccharide derivative represented by formula (1) [wherein X1 is a leaving group or a protected hydroxyl group, Y1 is a carboxyl group, a hydroxymethyl group, or a precursor group thereof, and R1 to R5 are independently hydrogen or a protecting group for a hydroxyl group]; a production method for the same; and a method for producing an oligomer of the disaccharide derivative using the disaccharide derivative as an intermediate.
Outline of related art and contending technology BACKGROUND ART
Glycosidic muscular dystrophy, a type of muscular dystrophy, is caused by the biosynthesis abnormalities of the glycoprotein dystroglycan, and is accompanied by central nervous disorders such as brain deformities and psychiatric retardation, including muscle cell injuries. Through recent studies, it has been found that abnormal sugar chains of dystroglycans present in muscle cells are responsible for abnormal sugar type muscular dystrophy. Among the saccharide chains of dystroglycans, the saccharide chain portion having affinity with laminin is referred to as a matrix glycan. Matriglycans connect muscle cells and basement membrane via laminin, and abnormal sugar chain muscular dystrophy develops due to biosynthesis abnormalities occurring in these sugar chains.
The overall structure of Ser / Thr and later sugars, including matrix glycans, has been investigated and found to be O-mannosylglycans in which matrix glycans are elongated via ribitol phosphate. A matrix glycan contains a saccharide chain having a repeating disaccharide including α-xylose and β-glucuronic acid in a non-reducing terminal portion thereof (Non-Patent Document 1), and it has been found that this saccharide chain having a repeating disaccharide is necessary for bonding to laminin (Non-Patent Document 2).
Improvements in genes that define carbohydrate biosynthases involved in the biosynthesis of O-mannosylglycans are promising for the treatment of abnormal carbohydrate muscular dystrophy. However, there are many types of genes involved, and when overexpressed, the pathology has also been reported to worsen, and gene therapy for abnormal carbohydrate muscular dystrophy is not easy.
