発明の概要(英語) The present invention addresses the issue of providing a recombinant phage that is highly safe and has excellent practicality and usefulness. Provided are: a recombinant bacteriophage that has lost proliferative capacity and can only infect once, as a result of having a bacteriophage genome having part of a virion-constituting gene deleted therefrom stored in the head thereof; and a preparation method therefor. Also provided are: a recombinant bacteriophage that has lost proliferative capacity and can only infect once, as a result of having a plasmid having a packaging site and coding a target gene stored in the head thereof; and a preparation method therefor.
従来技術、競合技術の概要(英語) BACKGROUND ART
Drug-resistant bacteria spread worldwide, while development of new antimicrobial agents is in progress. With respect to (set to which "set" is paid attention in such a situation, refer to, for example, Patent Document 1, Non-Patent Document 1, 2). Phage is a natural enemy virus infected with bacteria. Adhesion to (1) bacteria, injection of (2) phage genome, growth in (3) bacteria, (4) lysis, release of (5) progeny phages, and reinfection to (6) bacteria. Phages are used to treat bacterial infections. Since phage has extremely high host specificity, it is possible to sterilize almost only the target bacteria instead of indiscriminately sterilizing like antibacterial agents. Therefore, the bacterial flora which has been originally constructed can be treated without disturbing the bacterial flora.
