発明の概要(英語) The present invention can produce an L-amino acid oxidase at a low cost and in a large quantity through a high expression of a recombinant protein, and the like, and provides a novel L-amino acid oxidase (ArtLAAO) having a wide substrate selectivity. This novel L-amino acid oxidase is a protein comprising any one of amino acid sequences (1) to (4): (1) an amino acid sequence including an amino acid sequence represented by SEQ ID NO: 1, SEQ ID NO: 3 or SEQ ID NO; 5, and having an L-amino acid oxidase activity; (2) an amino acid sequence including an amino acid sequence represented by SEQ ID NO: 1, SEQ ID NO: 3 or SEQ ID NO: 5 with one or more amino acids deleted, substituted or added, and having an L-amino acid oxidase activity; (3) an amino acid sequence including an amino acid sequence having at least 90% amino acid identity with the amino acid sequence represented by SEQ ID NO: 1, and having an L-amino acid oxidase activity; and (4) an amino acid sequence including an amino acid sequence having at least 98% amino acid identity with the amino acid sequence represented by SEQ ID NO: 3 or SEQ ID NO: 5, and having an L-amino acid oxidase activity.
従来技術、競合技術の概要(英語) BACKGROUND ART
In recent years, attention has been paid to peptide drugs constituted by connecting a plurality of amino acids and derivatives thereof as new drugs. Peptide drugs have been reported to exhibit various functions due to a combination of DL-amino acids and derivatives thereof and a difference in steric structure. In order to synthesize peptide pharmaceuticals, it is necessary to obtain only one of D forms and L forms of amino acids, amino acid derivatives, or synthetic intermediates to be precursors thereof. This is due to the fact that most of the amino acids and their derivatives made by chemical synthesis are racemic. Therefore, dynamic optical resolution has been reported as a method for synthesizing only D amino acids or only L amino acids from racemic amino acids, amino acid derivatives or synthetic intermediates in high yield. The dynamic optical resolution method is a resolution method using an enzyme and can be carried out under normal pressure conditions, so that attention has been paid to today as an environmentally friendly clean method.
For the synthesis of L-amino acids by dynamic optical resolution, D-amino acid oxidase (DAAO) is generally used. Optical resolution of L-amino acids and derivatives thereof using DAAO has been reported in many documents as shown in Patent Document 1 and Non-Patent Documents 1 and 2 as an example, and has been widely used in industry.
On the other hand, in the synthesis of D-amino acids by dynamic optical resolution, L-amino acid oxidase (LAAO) is hardly used. When optically resolving only D isomers from racemic amino acids, amino acid derivatives, or synthetic intermediates by enzymatic methods, L-amino acid deaminase (LAAD) is widely used in industry, and a method of combining racemase with acylase or a method of combining racemase with amidase is used.
