発明の概要(英語) As a technique for detecting an influenza virus with improved accuracy, a method for detecting an influenza virus in a biological sample by using a first probe and a second probe is provided, wherein the first probe can be decomposed with each of a neuraminidase derived from an influenza virus and a neuraminidase derived from a bacterium to generate an optically detectable single and the second probe can be decomposed with a neuraminidase derived from a bacterium to generate an optically detectable single and cannot be decomposed with a neuraminidase derived from an influenza virus.
従来技術、競合技術の概要(英語) BACKGROUND ART
In recent years, see (Patent Document 1) has been developed a simple influenza virus test kit using immunochromatography. The method using immunochromatography is capable of detecting influenza virus within several minutes to several tens of minutes, and is therefore used for diagnosis and treatment of infection.
In addition, a technique for optically detecting influenza virus based on the reaction of neuraminidase (sialidase), which is an enzyme possessed by influenza virus, with a coloring substrate is conventionally known. Neuraminidase is an exo-glycolytic enzyme which liberates sialic acid from the non-reducing end of the sugar chain. Neuraminidases are present on the surface of influenza viruses and are involved in the proliferation of viruses. As the chromogenic substrate, for example, 4-methylumbelferyl-α-D-noiramic acid (4-acid: 4MU-NANA, see Patent Document 2), a chemiluminescent derivative of 4-alkoxy-N-acetylneuraminic acid or 4,7-dialkoxy-N-acetylneuraminic acid (, see Patent Document 3, and the like. For example, in a method using 4MU-NANA as a coloring substrate, 4-methylumbelliferone, which is a fluorescent substance, is produced by decomposition of 4MU-NANA by neuraminidase. The enzyme activity value of neuraminidase can be calculated on the basis of the fluorescence intensity of the generated 4-methylumbelferon, and the number of influenza virus particles can be quantified on the basis of the enzyme activity value.
Patent Document 4 discloses a kit for detecting whether influenza is human influenza or human pathogenic avian influenza in a human. The kit comprises a substrate comprising N-glycolide neuraminic (NeuGcα2-3Gal) acid bound to galactose and a substrate comprising N-acetylneuraminic acid (NeuAcα2-3Gal) bound to galactose. The 2 kinds of substrates are brought into contact with a clinical specimen, and when only the substrate based on NeuAcα 2-3Gal is cleaved, it is determined to be positive for human influenza virus, and when both the substrate based on NeuAcα 2-3Gal and the substrate based on NeuGcα 2-3Gal are cleaved, it is determined to be positive for human pathogenic avian influenza virus.
Patent Document 5 describes a method for detecting influenza virus by a digital method. In the digital method, an object to be detected is introduced into a large number of minimum spaces (, for example, minute droplets) so as to exist at a maximum of 1 molecules. Then, the object to be detected in each minimum space is detected, and its existence is digitized by 0 or 1. Thus, in the digital method, the detection object can be detected quantitatively with high sensitivity. Patent Document 6,7 describes a technique for encapsulating a substance in a minimum space (Enclose) and applicable to a digital method.
