発明の概要(英語) A cytotoxicity assay method that comprises: a step for co-culturing, in a buffer solution, cytotoxic T cells that are derived from a non-human animal having a T-cell receptor gene transferred thereinto with target cells that are labeled with a label and recognized by the cytotoxic T cells; and a step for measuring the label released from the target cells into the buffer solution, in which the amount of the label thus measured indicates the cytotoxic activity of the cytotoxic T cells, wherein the cytotoxic T cells are cells derived from a lymph node of the non-human animal. This cytotoxicity assay method is useful because a larger number of cytotoxic T cells can be produced thereby.
従来技術、競合技術の概要(英語) BACKGROUND ART
Cytotoxicity assays in which cytotoxic T cells are co-cultured with target cells and the number of damaged cells is detected are known as in vitro experimental methods for measuring the activity of cytotoxic T cells (see, e.g., NpL 1).
However, in conventional cytotoxicity assays, cytotoxic T cells must be pre-activated and cells that stimulate cytotoxic T cells must be provided in addition to cytotoxic T cells. Cells that stimulate cytotoxic T cells differ depending on the experimental system, but spleen cells of mice with different phenotypes of major histocompatibility complex (MHC), spleen cells of the same type of mouse immunized with an antigen, and the like are used, resulting in a very complex experimental system.
However, when spleen cells are used to stimulate cytotoxic T cells, the assay system contains cells other than cytotoxic T cells, and these cells affect the experimental system. Therefore, the inventors developed a simple cytotoxicity assay system that does not require stimulation of cytotoxic T cells (see NpL 2).
