発明の概要(英語) The present invention provides a peptide that disrupts the self-aggregation of an AtaA polypeptide or separates a bond between the AtaA polypeptide and another subject. A peptide found by the present inventors has the properties of disrupting the self-aggregation of an AtaA polypeptide or separating a bond between the AtaA polypeptide and another subject. A peptide according to the present disclosure includes amino acid sequences AVL, SVL, ATL, or functional equivalent sequences thereof. In one embodiment, a peptide according to the present disclosure has the ability to separate a bond between an AtaA and other molecules (for example, streptavidin and neutral avidin).
従来技術、競合技術の概要(英語) BACKGROUND ART
Bacteria of the genus Acinetobacter sp . Tol5 (Acinetobacter Tol5 strain) are non-pathogenic Gram-negative bacteria that have high cell self-aggregation and exhibit high adhesion to a variety of material surfaces, from a variety of hydrophobic plastic carriers to hydrophilic glass and metal surfaces. The present inventors discovered novel bacterio-nanofibers present in the bacterial cell surface layer as factors that provide such adhesion properties that are not reported by other microorganisms, and further identified novel proteins that constitute the nanofibers. This protein belongs to the trimeric autotransporter adherin (TAA) family of gram-negative bacteria, and the inventors named AtaA (NpL 1).
AtaA is extremely useful for immobilizing microorganisms and is expected to be applied to various bioprocesses (see, for example, Patent Documents 1 and 2). In addition, the present inventors reported that various functional peptides and proteins can be introduced into AtaA and presented on the surface of microorganisms in the previous patent application (Patent Document 3). Note that, in Patent Document 1, AtaA and a gene encoding the same (ataA gene) are referred to as AadA and aadA genes, respectively.
