発明の概要(英語) Provided is a technology for treating or preventing cancer by using GD2 as a molecule target. This GD2 binding molecule includes: a heavy chain variable region including a heavy chain CDR1 that has an amino acid sequence represented by SEQ ID NO: 1, a heavy chain CDR2 that has an amino acid sequence represented by SEQ ID NO: 2, and a heavy chain CDR3 that has an amino acid sequence represented by SEQ ID NO: 3; and/or a light chain variable region including a light chain CDR1 that has an amino acid sequence represented by SEQ ID NO: 9, a light chain CDR2 that has an amino acid sequence represented by SEQ ID NO: 10, and a light chain CDR3 that has an amino acid sequence represented by SEQ ID NO: 11.
従来技術、競合技術の概要(英語) BACKGROUND ART
A ganglioside is a type of glycolipid and is composed of a sugar chain portion and a lipid (ceramide: fatty acid+ long chain base). Gangliosides are synthesized by a series of enzymatic reactions and metabolized to end products. GD2 is synthesized from GD3 by GM2/GD2 synthase and further synthesized from GM1/GD1b / GA1 synthase to GD1b.
High expression of GD2 synthase and low expression of GD1b synthase in cancer cells results in high expression of GD2 on the cell surface. It is known that GD2 expressed on cells coexists with adhesion molecules such as Integrin and is involved in cell adhesion and signaling, and is involved in cancer growth and metastasis.
GD2 is known to be highly expressed in melanoma, neuroblastoma, glioblastoma, lung cancer, osteosarcoma, and leukemia. in normal tissues, expression is seen in neuronal cells and glial cells, but expression is low in normal tissues.
