In one embodiment, the present invention relates to: a method for producing a hollow collagen gel characterized by bringing a collagen solution filled in a material having a hole into contact with a gelling agent; or a hollow collagen gel tube produced using said method.
従来技術、競合技術の概要(英語) BACKGROUND ART
Collagen materials are widely used as surgical medical devices (TERUMO: Teradamis, GUNZE: Pernac). In recent years, attention has also been paid to the development of new medical technologies including regenerative medicine.
Examples of conventional collagen materials include sponge-like or membrane-like products obtained by vacuum drying. Recently, filamentous collagen that assists in the regeneration of nerves has been marketed (NIPRO: linave). The product is formed as a thread-like hydrogel by extruding collagen into an organic solvent. The thread-like collagen can then be used as a suture to a tissue transection, as a cell carrier seeded with cells, or as a derivative of cell migration in tissue regeneration. Since tissues are composed of epithelial cells and mesenchymal cells, regeneration thereof requires the simultaneous transplantation of different cells due to parenchyma cells (such as epithelial cells) and mesenchymal cells (such as vascular endothelium).
However, since the organic solvent remains with the method described above, it is a problem to solve the toxicity problem thereof. In particular, in the case of filamentous forms as the cell carrier required for regenerative medicine, it is not possible to encapsulate cells or granular drugs having a large diameter into the material. Furthermore, in the case of a thread-like shape, when two or more types of cells are seeded on the surface, cells with low growth potential are purged of cells with high growth potential, so use as a culture medium carrier is unsuitable.
On the other hand, the present inventors discovered that thread-like atelocollagen can be produced by subjecting a cylindrical hydrogel to ultraviolet irradiation, vitrifying, and rehydrating (Non-Patent Document 1: Biomater Sci . 2019 7 (1): 125-138).
In addition, techniques for manufacturing tubular tissue bodies include formation of high-density and highly oriented collagen tubular tissue bodies utilizing the contractility of smooth muscle cells and the possibility of application to ultra-small-bore artificial blood vessels (Non-Patent Document 2: Koda, Kaihiro Oka, Matsuda Warn-Artificial Organization , 1994), a collagen tube characterized by having a wall formed from a continuous, cylindrical, coaxial collagen film, and the like are also known (Patent Document 1: Japanese Unexamined Patent Application Publication No. 2009-540896). Furthermore, as a method for producing a collagen tube, Japanese Unexamined Patent Application Publication No. 48-033686 (Patent Document 2) discloses molding using a cylindrical nozzle, and Japanese Unexamined Patent Application Publication No. 2011-130995 (Patent Document 3) discloses a method for inserting and molding a rod-like member into a tube of a luminal collagen structure. Furthermore, Japanese Unexamined Patent Application Publication No. 04-035185 (Patent Document 4) discloses a molding method using a cylindrical nozzle, Japanese Unexamined Patent Application Publication No. GB1007978 (Patent Document 5) discloses an extruded collagen tube containing collagen fibrils, and Japanese Unexamined Patent Application Publication No. FR2902661 (Patent Document 6) discloses a method of winding a collagen film to form a tube.
