発明の概要(英語) Provided are: a novel membrane permeation enhancer which contains an amino acid-based surfactant and exhibits excellent safety; and a method for enhancing membrane permeation using this membrane permeation enhancer. The amino acid-based surfactant may be a compound represented by formula (1) or a salt thereof. In the formula, R1 denotes a hydrogen atom or a monovalent organic group, R2 denotes a hydrogen atom, -R4, -COR4 or -CH2CH(OH)R4, and R3 denotes -OH, -OR5, -NHR5 or -O(R6O)nH. One of R4 and R5 denotes an aliphatic hydrocarbon group having 6-20 carbon atoms, and the other denotes a hydrocarbon group which has 1-20 carbon atoms and which may include a hetero-atom. R6 denotes an alkylene group having 2-4 carbon atoms, and n denotes a number between 1 and 50. However, a case where R2 is a hydrogen atom and R3 is -OH or -O(R6O)nH is excluded.
従来技術、競合技術の概要(英語) BACKGROUND ART
A membrane such as a biomembrane has a role of separating the inside of the membrane from the outside so that a normal biofunction or the like can be exhibited inside the membrane surrounded by the membrane. Furthermore, these membranes do not simply isolate the inside from the outside. for example, in a case of cell membranes, these membranes are provided with devices for exchanging substances, information, and energy with the outside of the cells and performing various metabolic reactions. For example, specific transporters and receptors exist in the cell membrane, and these functions selectively incorporate necessary substances and discharge various metabolites.
In recent years, water-soluble membrane-permeable substances that permeate through membranes such as cell membranes have become known regardless of these specific transporters and receptors, and application to drug delivery and the like is anticipated.
However, in order to control the membrane permeability of these membrane permeable substances, there are a large number of considerations to be considered, such as concentration, application time, structure of the membrane permeable substance itself, and the like, and there is a problem in that it is not easy to change the membrane permeability according to the applied medicament.
As a means for solving such problems, the present inventors reported a method for controlling the membrane permeability of a membrane permeable substance by reinforcing or inhibiting the action of the membrane permeable substance to change the curvature of the membrane (see, for example, Patent Document 1). This report actually demonstrates that the membrane permeability of the Arg oligomer is improved using a method of changing the osmotic pressure between solvents separated by the membrane and a method of adding 1,3-butanediol and sodium thiocyanate, which are known to have an action of changing the curvature of the membrane to a convex shape.
However, the method for changing osmotic pressure is not realistic in applications to living organisms. In addition, considering the influence on cells or the like of the effective amount of 1,3-butanediol or sodium thiocyanate in the report described above, a method capable of safely promoting permeation of a membrane permeable substance at a lower concentration is desired.
