Title MESOTHELIOMA-SPECIFIC PROMOTER AND USE THEREOF
Abstract Disclosed is a promoter which has a transcriptional activity specific to mesothelioma and exhibits a low transcriptional activity on other cancer cells and normal cells including a mesothelium. Also disclosed is use of the promoter, specifically a vector for use in gene therapy or a therapeutic agent for mesothelioma, which comprises the promoter. Specifically disclosed is a promoter derived from a gene of CRI1 which is one of the markers for mesothelioma. Also specifically disclosed is a vector having a cell death-inducing gene or a cell lysis-inducing gene as a transgene and carrying a promoter derived from CRI1 gene upstream from the transgene. By using the vector, it becomes possible to elicit a cell death- or cell lysis-inducing activity in a mesothelioma-specific manner. Each of the vector for use in gene therapy and the therapeutic agent for mesothelioma comprises a viral vector carrying the promoter derived from CRI1 gene.

【請求項1】CRI1(CREBBP/EP300 inhibitory protein 1)遺伝子由来のプロモータであり、中皮腫特異的に転写活性を示すことを特徴とする新規プロモータ。

【請求項2】CRI1遺伝子由来のプロモータの配列が、CRI1遺伝子の-2586~+84の領域から選択される配列である、請求項1に記載の新規プロモータ。

【請求項3】CRI1遺伝子由来のプロモータの配列が、配列表の配列番号1~11に記載のいずれかである、請求項1または2に記載の新規プロモータ。

【請求項4】請求項1~3のいずれか1に記載の新規プロモータを搭載してなるウイルスベクター。

【請求項5】ウイルスベクターが、アデノウイルスベクターである請求項4に記載のウイルスベクター。

【請求項6】アデノウイルスが、制限増殖型アデノウイルスである請求項5に記載のウイルスベクター。

【請求項7】さらに、細胞死誘導遺伝子および/または細胞融解誘導遺伝子をプロモータの下流に搭載してなる請求項4~6のいずれか1に記載のウイルスベクター。

【請求項8】請求項4~7のいずれか1に記載のウイルスベクターからなる遺伝子治療型中皮腫治療用ベクター。

【請求項9】請求項8に記載の遺伝子治療型中皮腫治療用ベクターを含む中皮腫治療剤。

【請求項10】さらに、マーカー遺伝子をプロモータの下流に搭載してなる請求項4~6のいずれか1に記載のウイルスベクター。

【請求項11】マーカー遺伝子が、蛍光タンパク質発現遺伝子である請求項10に記載のウイルスベクター。

【請求項12】請求項10または11に記載のウイルスベクターからなる中皮腫検査用ウイルスベクター。

【請求項13】請求項12に記載の中皮腫診断用ウイルスベクターを用い、マーカーの発現の有無を観察することによる中皮腫の検査方法。
