Title AGENT FOR IMPROVING GENE TRANSFER EFFICIENCY TO MAMMALIAN CELLS
Abstract Provided is an agent for improving gene transfer efficiency to mammalian cells, a method of improving gene transfer efficiency to mammalian cells, and a transformation method for mammalian cells. The method is characterized in that tRNA is used in conjunction with a lipofection reagent. Preferably, the tRNA concentration in a lipofection solution falls within the range of 3 to 50 .mu.g/mL, and the concentration within a culture solution is approximately 1/10. More preferably, tRNA and PEG can be used in conjunction with a lipofection reagent. As a consequence, it is possible to achieve a high gene transfer efficiency to mammalian cells.

【請求項1】tRNAを含有し、リポフェクション試薬と併用することを特徴とする、哺乳動物細胞への遺伝子導入効率の向上剤。

【請求項2】tRNA及びリポフェクション試薬を含有することを特徴とする、哺乳動物細胞への遺伝子導入効率の向上剤。

【請求項3】リポフェクション溶液中のtRNA濃度が3~50μg/mLの範囲内となるように用いることを特徴とする請求項1又は2に記載の向上剤。

【請求項4】さらにポリエチレングリコールを併用することを特徴とする請求項1~3のいずれかに記載の向上剤。

【請求項5】ポリエチレングリコールの分子量が、2000~6000の範囲内であることを特徴とする請求項1~4のいずれかに記載の向上剤。

【請求項6】ポリエチレングリコールの分子量が、3000~5000の範囲内であることを特徴とする請求項1~4のいずれかに記載の向上剤。

【請求項7】哺乳動物が、ヒト、マウス、サルから選ばれる哺乳動物であることを特徴とする請求項1~6のいずれかに記載の向上剤。

【請求項8】tRNAをリポフェクション試薬と併用することを特徴とする、哺乳動物細胞への遺伝子導入効率の向上方法。

【請求項9】tRNAをリポフェクション試薬と併用することにより、目的遺伝子を哺乳動物細胞へリポフェクションすることを特徴とする哺乳動物細胞の形質転換方法。
