The object is to provide: a therapeutic or prophylactic agent for metabolic syndrome, which has improved stability in a living body; and others.
The therapeutic or prophylactic agent for metabolic syndrome comprises, as an active ingredient, DNA that encodes receptor activity-modifying protein (RAMP) 2 and is selected from the items (a) to (d) below or a polypeptide encoded by the DNA: (a) DNA which has the nucleotide sequence depicted in SEQ ID NO:1; (b) DNA which has a nucleotide sequence capable of hybridizing with the nucleotide sequence depicted in SEQ ID NO:1 under stringent conditions; (c) DNA which has a nucleotide sequence encoding an amino acid sequence having the substitution, deletion and/or addition of one or more amino acid residues in the amino acid sequence depicted in SEQ ID NO:2; and (d) DNA which has a nucleotide sequence having a 90% or more homology with the nucleotide sequence depicted in SEQ ID NO:1.
Scope of claims [claim1]
1. An agent for use in the therapy or prophylaxis of metabolic syndrome wherein the agent comprises a DNA described in any one of (a) to (c) below, encoding receptor activity modifying 2 (RAMP2) protein, or a polypeptide encoded by the DNA; (a) a DNA having a base sequence described in SEQ.
ID. No. 1; (b) a DNA having a base sequence which is capable of hybridizing under stringent conditions with the base sequence described in SEQ.
ID. No. 1; (c) a DNA consisting of a base sequence having a homology of 90% or more with the base sequence described in SEQ.

2. An in vitro test method for testing the contraction of metabolic syndrome or testing the propensity to contract metabolic syndrome comprising: an extraction step of extracting DNA from a cell from a test subject; an amplification step of carrying out a polymerase chain reaction using a primer which is capable of selectively amplifying a DNA consisting of the base sequence described in SEQ.
ID. No. 3, with the extracted DNA as a template; a determination step of determining a base sequence of the amplified DNA; a comparison step of comparing the determined base sequence with the base sequence described in SEQ.

No. 3; and, wherein differences are noted, it is judged whether or not the symptoms relating to metabolic syndrome are improved or their worsening is suppressed, when the DNA amplified in the amplification step is introduced into a cell, tissue, or organ where the RAMP2 has been mutated to remove its function or knocked out.
