Disclosed is an RNAi regulation system utilizing an RNA-protein interaction motif. Specifically disclosed is shRNA which comprises a guide strand comprising a sequence complementary to a target sequence, a passenger strand capable of being annealed to the guide strand to form a double strand, and a linker strand capable of linking the guide strand to the passenger strand, wherein the linker strand comprises an RNP-derived protein-binding motif sequence. Also disclosed is an RNAi regulation system which comprises the shRNA and an RNP-derived protein capable of binding specifically to a protein-binding motif sequence in the shRNA.
