発明の概要(英語) Provided are: a variant reverse transcriptase which has a high heat stability and a high versatility; a nucleic acid thereof; a method for producing the variant reverse transcriptase; a highly versatile reverse transcription kit; a highly versatile detection kit; a method for improving the heat stability of a nucleic acid enzyme whereby the heat stability of the nucleic acid enzyme can be highly improved; and a reverse transcription method whereby reverse transcription can be efficiently conducted. An amino acid residue in a nucleic acid interaction domain of a wild type enzyme is substituted by a positively charged amino acid residue or a nonpolar amino acid residue to form a nucleic acid interaction domain having a larger positive net charge than the nucleic acid interaction domain of the wild type enzyme.
従来技術、競合技術の概要(英語) BACKGROUND ART
Reverse transcriptase, in general, active RNA as a template to synthesize cDNA (hereinafter, 'RNA-dependent DNA polymerase activity' that) and, as a template to synthesize DNA DNA activity (hereinafter, 'DNA-dependent DNA polymerase activity' that) and, RNA: the decomposition of chain RNA in the hybrid DNA (hereinafter, 'RNase H activity' that) has.
