Abstract

It is intended to obtain a cassette for fixing, embedding and slicing a biological tissue by forming a container made of a sliceable material, sampling a biological tissue and packing it in the container, and then conducting a whole process of making a preparation involving fixation and slicing without using any other container so that accurate tissue inspection data of the biological tissue can be obtained while simplifying the procedures, minimizing the risk of crushing the tissue and avoiding a mistake such as misidentification of preparations, and a method of fixing and embedding a biological tissue to form a block followed by fixation, embedding and slicing with the use of this cassette. A cassette (10A) for fixing, embedding and slicing a biological tissue comprising a base sheet (11') which is made of a sliceable material sheet having a strength of a definite level or higher and a cover sheet (12'), wherein the base sheet (11') has a groove (13) as a container for containing a biological tissue, and the cover sheet (12') is formed by half folding a continuous plastic sheet made of the same material as the base sheet (11') at the folding part (14) to unify it and the base sheet (11'), thereby giving an open/close cover sheet (12'). A biological tissue is tightly adhered to a site in the bottom of the cover sheet (12') fitting for the bottom face structure of the base sheet (11') by using a sponge material (13a) for tightly adhering and holding the tissue. After dehydration and penetration, the tissue is embedded into paraffin employed as an embedding material. Then, a block for slicing including an embedding frame is formed and the block is sliced with a microtome. Thus an excellent biological tissue preparation can be formed.
