発明の概要(英語) Provided is a peptide that specifically binds to IgY. A peptide characterized by: containing an amino acid sequence that consists of 17 amino acid residues and is represented by a formula: (X1-3)-C-(X5)-W-(X7-11)-W-(X13)-C-(X15-17) [wherein X1 and X3, among X1-3, and X15-17 independently represent an arbitrary amino acid residue other than a cysteine residue; X2 represents an arbitrary amino acid residue that may be a cysteine residue; C represents a cysteine residue; X5 represents an amino acid residue selected from among T, D, V, S and E; W represents a tryptophan residue; X7-11 represents an amino acid sequence (S, T, R or N)-(S, A, K or R)-(I, A, V, T or G)-(V, Y, T, R or S)-(G or D); and X13 represents an amino acid residue selected from among V, L, D, S, M, T and R]; a disulfide bond being formed between cysteine residues at the 4- and 14-positions; and being capable of binding to tri-IgY. 従来技術、競合技術の概要(英語) BACKGROUND ART
The current, as a reagent for the study of clinical diagnostics and the antibodies of the, or a rabbit polyclonal antibody with murine monoclonal antibody and are frequently used, has a large market. On the other hand, chicken IgY antibodies, as well as establishing the method for fabricating the immune specific antibody, and reworking the cost of the test animal, the animal antibody in addition to the low cross-reactivity, non-invasively from egg yolk antibodies IgY merit attention as purification, as reagents are expected as a new antibody.
As a method of purifying a IgY from egg yolk of, several methods have been reported (non-patent document 1). Is in the egg yolk, phospholipid containing a large amount of insoluble and Lipoprotein and, when the affinity purification of these thus hindering, this PEG, such as Pectin removed by precipitation method using reported protocol (non-patent document 2). Commercial IgY purification kits using the same principle and also, the current, are mainly, and is EGGstract (Promega) Eggcellent (Thermo Pierce) have been frequently used. Recently, Sock Hwee Tan et al., κ-carrageenan and plant gums pectin was used for precipitation, also commercially available, which is not inferior in purity and yield reported purification protocol (non-patent document 3). However, in such a method also, the purity of the purified IgY is about 50-80%, for the preparation of more highly purified IgY, is essential for the use of affinity columns.
However, for example, in human IgG such as Protein A, specific ligand for the purification of IgY is not reported, synthetic ligands for purification but has been reported (non-patent document 4, manufactured by GE Healthcare 2-mercaptopyridine or immobilized column: HiTrap IgY Purification HP), the purity of the purified IgY antibody (about 70%) from the viewpoint of, needs to be evened out to be insufficient.
The present inventors have, heretofore, were constructed by a phage display system using T7, specific for human IgG or human IgA isolated peptides that bind to, the peptide is immobilized on the affinity column purification using antibodies has been reported (Patent Document 1, 2).
However, for any peptide IgY binding, this was not enough to report.
