Title CELL STIMULATION DEVICE meetings
Abstract PROBLEM TO BE SOLVED: To provide a cell stimulation device that can give stimulation meeting the purpose or use selectively into an arbitrarily intracellular constituents.
SOLUTION: The device is equipped with a sine wave signal generator 10 and a pair of parallel flat plate electrodes 12, 13 and burst high-frequency electric field is generated from the pair electrodes 12, 13 that have, as the fundamental frequency, the value equal to the inverse of the relaxation, time of the dielectric material included in the element arbitrarily selected among the intracellular constituents. The burst high-frequency electric field means an AC electric field generated continuously and periodically for a prescribed period and most of the input power are concentrated in the area of the fundamental frequency band.

従来より、矩形状のパルス電界により細胞膜に小孔を開け、その小孔から細胞内に遺伝子を導入するエレクトロポレーションが知られている。このエレクトロポレーションは、導電性の細胞質が誘電性の細胞膜で覆われた構造の細胞と、一対の平行平板電極とを導電性の懸濁液中に入れた状態で、その一対の電極から細胞および懸濁液にパルス電界を印加して細胞の細胞膜に電気的な刺激を与えるものである。細胞膜に与えられる電気的な刺激は、電界の印加によって懸濁液中を流れる電流が細胞膜の表面に電荷を帯電させ、細胞膜に電気的および機械的なストレスを生じさせることにより起こるものであり、そのストレスによって細胞膜を一時的に破壊して細胞膜に小孔を形成する。細胞膜を刺激するために印加されるパルス電界は、一般に、パルス幅が数十μs以上と長く、振幅が数十Vrms程度と低いものである。それは、細胞膜を電気的に刺激するのに要する時間（緩和時間）が数十μs程度と長いからであり、このように長時間に渡って電界を印加しても細胞が死なないようにするには振幅を低く抑える必要があるからである。



このように、エレクトロポレーションでは、パルス幅が数十μs程度と長く、振幅が数Vrms程度と低いパルス電界が長らく用いられてきた。しかし、近年、緩和時間以下のパルス幅のパルス電界を細胞に加えると、細胞膜を通して懸濁液および細胞質に電界の大きさに応じた電流が流れることがわかってきたことから、その応用研究が活発に行われている。非特許文献1では、それを利用して、細胞膜を破壊しないで細胞内の構成要素、例えば核膜に電界を作用させる技術が開示されている。

【請求項1】
 
細胞内構成要素のうち任意に選択した要素に含まれるDNAの緩和時間の逆数と等しい値の周波数を基本周波数とするバースト高周波電界を発生する電界発生手段
を備えたことを特徴とする細胞刺激装置。

【請求項2】
 
前記基本周波数は、1MHz以上1GHz以下である
ことを特徴とする請求項1に記載の細胞刺激装置。

【請求項3】
 
前記バースト高周波電界の電界強度と印加時間との積が所定の範囲内となるように、前記バースト高周波電界の電界強度および印加時間がそれぞれ設定される
ことを特徴とする請求項1に記載の細胞刺激装置。
