Title	METHOD FOR PRODUCING NEW ANTIBODY ENZYME AND THE NEW ANTIBODY ENZYME
Abstract	PROBLEM TO BE SOLVED: To provide a method for efficiently producing an antibody enzyme together having high molecule-recognizing ability as an antibody and activity as an enzyme, to provide the new antibody enzyme obtained by the method, and to provide another antibody enzyme applicable to the method and having a three-dimensionally specific structure. SOLUTION: This method for producing the antibody enzyme includes an antibody structure-analyzing process for confirming whether a triadic structure of catalyst residues in which a serine residue, an aspartic acid residue, and a histidine residue or a glutamic acid residue are arranged in a neighboring space in a three-dimensional structural manner exists or not in a three-dimensional structure of the antibody estimated based on its amino acid sequence. Because the triadic structure of the catalyst residues is such a structure as specific to the antibody enzyme, the antibody enzyme is efficiently screened thereby.
Outline of related art and contending technology	(In Japanese) 近年、酵素様活性をもつ抗体、即ち、抗体酵素について、種々の報告がなされている。例えば、S.Paulらは、自己免疫疾患患者の分離精製した自己抗体がVIP（神経ペプチド：Vasoactive Intestinal Peptide）を分解する活性を有することを報告している（非特許文献1）。また、Gabibovらは、SLE（Systemic lupus erythematosus）等の自己免疫疾患や免疫細胞増殖性疾患患者から分離した自己抗体にDNAを酵素的に分解する抗体が存在することを報告している（非特許文献2および非特許文献3）。また、本発明者らは、AIDS（後天性免疫不全症候群）の原因となるエイズウイルス（HIV）の外膜タンパク質gp41の不変領域を抗原とするモノクローナル抗体を取得し、このモノクローナル抗体の機能を詳細に解析した。その結果、このモノクローナル抗体の軽鎖領域は、HIVの外膜タンパク質gp41を特異的に分解する、非常に高い活性を有することを報告している（非特許文献4）。このように抗体酵素は、抗体の高い分子認識能と酵素活性とを併せ持つため、医療、化学工業、食品工業等といった、多くの面で応用が期待されている。これまで、このような抗体酵素を作製・取得する方法としては、基底状態にあるペプチド、あるいはタンパク質を動物等に免疫し、得られた抗体の全てについて酵素活性を測定して、目的の酵素活性を有する抗体だけを選択して取得するという方法しか知られていなかった。【非特許文献1】 Paul, S., et al., Science, 244: 1158, 1989. 【非特許文献2】 Shuster, A.M., et al., Science, 256: 665, 1992., 【非特許文献3】 Gabibov, A.G., et al., Appl. Field of industrial application	(In Japanese) 本発明は、抗体の高い分子認識能と酵素活性とを併せ持つ抗体酵素を高い効率で生産する新規な抗体酵素の生産方法と、この生産方法により生産される抗体酵素と、この生産方法に応用できる立体的に特異的な構造を有する抗体酵素とに関するものである。
Scope of claims	(In Japanese) 【請求項1】ヘリコバクター・ピロリ菌のウレアーゼに対する抗体断片であり、かつ、ペプチドの分解酵素として作用する抗体酵素であって、配列番号16に示すアミノ酸配列を含んでなることを特徴とする抗体酵素。【請求項2】請求項1に記載の抗体酵素をコードする、遺伝子。【請求項3】配列番号17に示す塩基配列からなることを特徴とする請求項2に記載の遺伝子。【請求項4】請求項2または3に記載の遺伝子を含む、ベクター。【請求項5】請求項2または3に記載の遺伝子が導入されている、ホスト細胞。
